Khoo Hooi-Hye was educated at the Penang Free School, where started playing lawn tennis at the age of 15. His first victory in a tournament came 1918 at the Penang Chinese Recreation Club championship, the first of several to come. In 1922 Khoo won the covered court championship at Singapore. In the following year he won both, the Singapore and Malayan titles for the first time. In 1924, he went to Europe to become one of the earliest Chinese to compete at Wimbledon. Khoo Hooi-Hye was also entered in the Olympic Games lawn tennis tournament, but did not start. After his return to Malaya in 1925, Khoo Hooi-Hye dominated the local lawn tennis scene, winning five more Singapore titles and four more Malayan titles until 1929.
In 1927, Khoo represented China at the Far Eastern Games, winning the event together with compatriot Lin Baohua. He competed in the 1930 Far Eastern Games again. For the next ten years Khoo Hooi-Hye lived in Shanghai successfully playing national and international tournaments in South-East Asia. He returned to Malaya in 1936.
