A middle defender, Karel Knesl played football at the 1964 Tokyo Olympics for Czechoslovakia, helping them win a silver medal. He started his youth career with TJ Gottwaldov in 1955, moving to Dukla Praha in 1962. He played with them from 1962-65 and then joined SK Slavia Praha (1966-70), before finishing his career with Škoda Plzeň in 1970/71. He had his greatest success with Dukla, winning three Czechoslovak league titles, three American Cup victories, and winning the Czechoslovak Cup.
